This blog post delves into our platforms, showcasing their potential for reshaping the landscape of healthcare.

A. "Plug and Play" antibody-mimetic molecular assembly platform
Our first platform is based on a safe, widely used protein nanocage called ferritin. It is a hollow spherical-shaped protein made of 24 subunits. Using our proprietary engineering methodology, we converted the genetic code of each subunit into that of an antibody-mimetic precursor comprising two constant domains linked by a linker peptide and a variable N-terminal domain inserted into the constant domain via a peptide arm. The linker peptide between the two constant domains encodes the sequence recognized by a site-specific protease. Consequently, the genetic code is used to express each precursor, and protease cleavage of the precursors yields nanocages decorated with multiple variable domains.

The variable domain can be rapidly changed and replaced by targeting ligands, antibody domains, antigens, or therapeutic proteins. The peptide arm could be designed for site-specific small-molecule or protein conjugation, and the C-terminus can be modified to include therapeutic peptides. The protease cleavage assembly can be used to encapsulate hydrophilic or hydrophobic drugs.

B. "Plug and Play" restriction factor enzymes for antiviral therapy
By engineering human enzymes, we have created humanized multidomain viral restriction factor enzymes. These enzymes are being tested and developed for gene therapy to create a synthetic innate immune system that can prevent or treat viral infections.
